ImagePro>Cool Snap Pro & IPP for semi-quantitative densitometry



To All,

I am interested in relative differences between staining
(immunohistochemistry) of rat brain sections. I would like to use
ImageProPlus to do this, and we have a Cool Snap-Pro cf monochrome camera.

One question is whether anyone knows how to determine whether this camera
has a linear response. I have read that this is important for
semi-quantitative densitometry.

My next question pertains to calibration. I believe I should select Std.
Optical Density for calibration. I wondered if I could calibrate using one
image that is as dark as possible (e.g., a solid object placed on top of a
slide) and one image that is as light as possible (e.g., a blank slide).
Would this be appropriate?
