I do not know whether Manual Tag analysis utilizes circular AOIs. However, trying to overcome the problems of Tags with a set of circular AOIs, I found the same kinds of problems: circular AOIs give incorrect (unexplainable) averaging with 12 and 16 bit images, independently from the pixel sampled and in absence of intensity calibrations; in addition there is an imperfect matching between the circular AOI boundaries and the pixels sampled.
If I insist on this problem is not to criticize IPP, with I consider a powerful, versatile and invaluable program, but solely to signal a bug.
